SODIUM REAGENTS

Enzymatic method
FIXED TIME

PRINCIPLE

Sodium measurements are used to monitor electrolyte balance in the diagnosis and treatment of disease conditions characterised by low or high blood sodium levels. Sodium is determined enzymatically via sodium-dependent β- galactosidase activity with ONPG as substrate. The absorbance at 405 nm of the product O-nitrophenyl is proportional to the sodium
concentration.

REAGENT COMPOSITION

R1 Good’s buffer (pH 8.5), β-D-galactosidase (<8 U/mL), Cryptand (>0.4 mM), Proclin 300 (0.02%). Good’s buffer (pH 6.5), ONPG (>0.5 mM), Proclin 300 (0.02%).

R2 CAL Sodium / Potassium standard. Sodium (Na+ ) 160 mmol/L. / Potassium 6.0 mmol/L.

STORAGE AND STABILITY

Store at 2-8ºC. The Reagents are stable until the expiry date stated on the label.

REAGENT PREPARATION
Ready-to-use.
SAMPLES
Serum. Sodium is stable for at least 24 hours at room temperature
and 2 weeks when refrigerated.

MATERIALS REQUIRED

  • Photometer or spectrophotometer with a thermostatic cell compartment set at 37ºC, capable of reading at 405 nm.
  • Pipettes to measure reagent and samples
